The aim of this study was to evaluate the bioactive properties of ethyl acetate crude extracts of Streptomyces sp. PERM2 and PFK4 with a view to assess their therapeutic potential. The antimicrobial activities of the crude extracts were assayed against fungal and bacterial pathogens using microdilution technique. The antioxidant activity of crude extracts (DPPH, FRAP and Metal chelating activity) were also evaluated. The antiproliferative effect of extracts was evaluated on the model of benign prostate hyperplasia (BPH) induce by the testosterone in Wistar male rats. The crude extracts chemical constitutes were determined by the LC-MS/MS. The PERM2 and PFK4 extracts presented minimal inhibitor concentration (MIC) and minimal microbicide concentration (MMC) values ranging from 31.25 µg/mL to 2000 µg/mL. The amount of total phenolic compounds was of 16.75 µg and 19.75 µg AAE/mg in PERM2 and PFK4 extracts respectively. The total antioxidant capacity was 0.69 and 0.90 µg AAE/g in PERM2 and PFK4 extracts respectively. The extracts of PERM2 and PFK4 have a dose-dependent reducing power [0.408 (PERM2), 0.597 (PFK4)] and antiradical activity [46.41% (PERM2), 74.94% (PFK4)]. The PFK4 extract tested at 150 mg/kg significantly reduced the volume of prostate by 48.45%, prostatic epithelium length (54%), proteins level (8.88%), and inhibited the development of HBP ranging from 41.11% (prostatic index) to 44.76 % (prostate weight), compared to the negative control. The LC-MS/MS analysis revealed the presence of about twenty two and twenty five bioactive metabolites in PERM2 and PFK4 crude extracts and those identified were 3.30-O-diméthylellagic acid, amicoumacin A, fongichromin, N-acetyl D-L phenylalanine and rapamycin. The results revealed that the ethyl acetate extracts from Streptomyces sp. PERM2 and PFK4 possess a wide spectrum of antimicrobial, antioxidant and antiproliferative activities which affords the production of significant bioactive metabolites as potential pharmacological agents.
